灵芝化学成分的HPLC-QTOF MS分析

摘要(Abstract):

目的使用高效液相色谱-四级杆飞行时间质谱联用技术(HPLC-QTOF MS)同时对灵芝中的核苷类、三萜类和甾醇类成分进行分析和鉴定。方法采用甲醇加热回流提取制备样品;色谱柱为Agilent Polaris C18-A(150 mm×4.6 mm,5μm),流动相为0.02%甲酸水-0.02%甲酸乙腈,梯度洗脱,进样量为2μL,检测波长核苷类和三萜类成分为260 nm,甾醇类成分为275 nm;ESI源,正离子模式下采集一、二级质谱数据,通过与文献或对照品对照,确定化合物成分。结果共鉴定出30个成分,包括3个核苷类成分,26个三萜类成分,1个甾醇类成分。结论本研究建立的HPLC-QTOF MS方法,可以同时对灵芝中的强极性到弱极性成分进行综合分析,为灵芝药材质量控制提供了参考。
OBJECTIVE To develop a high performance liquid chromatography-quadrupole time of flight mass spectrometry(HPLC-QTOF MS) method for simultaneous analysis of nucleosides,triterpenoids and sterols in ganoderma. RESULTS The sample solution was prepared with methanol by heat reflux extraction. The separation was performed on a Agilent Polaris C_(18)-A column(150 mm×4. 6 mm,5 μm) with gradient elution by water and acetonitrile containing 0. 02% formic acid. The injection volume was 2 μL.Nucleosides and triterpenoids were detected at 260 nm,and sterols were detected at 275 nm. ESI source was used,MS and MS/MS data were acquired in positive mode. Compounds in ganoderma were identified by comparing with the reference substance and the literature data. RESULTS A total of 30 compounds,including 3 nucleosides,26 triterpenoids and 1 sterol were identified. CONCLUSION Simultaneous analysis of polar and less polar compounds in ganoderma by HPLC-QTOF MS is established,which is suitable for quality evaluation of ganoderma.
