狼疮方颗粒对主动型Heymann肾炎大鼠模型的药效学研究

摘要(Abstract):

目的研究狼疮方颗粒对主动型Heymann肾炎(active heymann nephropathy,AHN)大鼠模型的药效学作用,为狼疮方颗粒临床应用提供实验依据。方法建立AHN大鼠模型,将实验动物随机分为正常对照组、模型对照组、阳性对照组和实验组(高、中、低剂量组),检测给药后各组动物模型中尿液的尿蛋白含量以及血清抗双链DNA抗体(ds-DNA)、抗核抗体(ANA)、肌酐(Cr)、尿素氮(BUN)、总补体活性(CH50)、免疫球蛋白G(Ig G)等的水平,探讨狼疮方颗粒治疗狼疮性肾炎的药效。结果与模型对照组相比,狼疮方颗粒能显著降低AHN大鼠尿蛋白、Cr和Ig G水平(P<0.05),狼疮方高剂量组能显著降低BUN的水平(P<0.01);醋酸泼尼松、狼疮方颗粒对AHN大鼠ds-DNA、ANA、血清CH50水平均无显著影响(P>0.05)。结论狼疮方颗粒对AHN疗效明确且对肾脏具有保护作用,可能与减少肾脏免疫复合物的沉积有关。
OBJECTIVE To investigate the pharmacodynamical effect of Langchuangfang granule on active heymann nephropathy( AHN) model in rats,providing experimental evidence for clinical application of Langchuangfang granule. METHODS AHN rat model was firstly established and rats were randomly categorized into normal control group,model control group,positive control group and experimental group( high,medium and low-dose group). Then the level of indicators,including urinary protein,ds-DNA,ANA,Cr,BUN,CH50 and IgG were determined after administration in different groups. At last the therapeutic effect of Langchuangfang granule on lupus nephritis( LN) was discussed. RESULTS In comparison with model control group,Langchuangfang granule in all doses could significantly decrease the level of urinary protein,Cr and IgG( P < 0. 05),and high-dose Langchuangfang granule group could significantly reduce the level of BUN( P<0.01). Prednisone acetate,Langchuangfang granule had no significant influence on the level of ds-DNA,ANA and CH50( P>0.05). CONCLUSION Langchuangfang granule has a protective influence on kidney,which possibly relates to decreased deposition of immune complex on it,confirming the efficacy of Langchuangfang granule in treating AHN.
