岭南饮片制枳壳不同化学部位对功能性消化不良大鼠的影响

摘要(Abstract):

目的探究岭南特色饮片制枳壳不同化学部位对功能性消化不良(functional dyspepsia, FD)大鼠胃肠动力和脑肠肽含量的影响。方法将48只雄性SD大鼠随机分为空白组、模型组、莫沙必利组、石油醚-乙酸乙酯组、正丁醇组、萃余组、挥发油组和总黄酮组,除正常组外,其他实验组大鼠采用"夹尾+不规则饮食"法制备FD模型。持续给药2周后,测定大鼠胃残留率和小肠推进率,酶联免疫吸附法(ELISA)检测大鼠血清中生长抑素(SS)、神经降压素(NT)、胃泌素(GAS)、胃动素(MTL)水平。结果与模型组相比,挥发油部位可降低大鼠胃残留率(P<0.05),总黄酮部位可升高大鼠的小肠推进率(P<0.01);正丁醇部位及总黄酮部位可降低FD大鼠血清中SS及NT的含量(P<0.01),萃余部位和总黄酮部位可升高GAS含量(P<0.05),石油醚-乙酸乙酯部位及总黄酮部位可升高MTL含量(P<0.05)。结论制枳壳可改善功能性消化不良相关症状,不同化学部位的作用机制不同,这与胃肠道动力改变及脑肠肽SS、NT、GAS和MTL的分泌异常有关。
OBJECTIVE To explore the effects of different chemical parts of processed Fructus Aurantii from Lingnan characteristic decoction pieces on gastrointestinal motility and brain intestinal peptide content in rats with functional dyspepsia(FD). METHODS 48 male SD rats were randomly divided into control group, model group, Mosapride group, petroleum ether ethyl acetate group, n-butanol group, raffinate group, volatile oil group and total flavonoids group.Except for the control group, the FD model was prepared by "tail clamping+irregular diet" method to other groups.After continuous administration for 2 weeks, the gastric residue rate and small intestinal propulsion rate were measured.The levels of somatostatin(SS),neurotensin(NT),gastrin(GAS) and motilin(MTL) in serum were measured by enzyme-linked immunosorbent assay(ELISA). RESULTS Compared with the model group, the fraction of volatile oil could reduce the gastric residue rate(P<0.05).And the fraction of total flavonoids could increase the intestinal propulsion rate(P<0.01).N-butanol and total flavonoids could reduce the contents of SS and NT in serum of FD rats(P<0.01).The extract part and total flavonoids could increase the content of GAS(P<0.05).The petroleum ether ethyl acetate part and total flavonoids could increase the content of MTL(P<0.05). CONCLUSION Processed Fructus Aurantii can improve the symptoms of functional dyspepsia.The mechanism of action of different chemical parts are different, which are related to the changes of gastrointestinal motility and the abnormal secretion of brain intestinal peptides SS,NT,GAS and MTL.
