PDGF-BB对大鼠肝星状细胞T6增殖及细胞外基质的影响

摘要(Abstract):

目的研究血小板衍生生长因子-BB(PDGF-BB)在不同条件下对大鼠肝星状细胞系T6(HSC-T6)增殖及表达细胞外基质的影响。方法不同浓度(5、10、20、40 ng/mL)的PDGF-BB在不同的孵育时间(3、6、12、24、36 h)时刺激HSC-T6细胞,噻唑蓝(MTT)法观察细胞增殖作用,RT-PCR检测CollagenⅠ、TGF-β1、TIMP-1和MMP-2 mRNA表达量。结果 PDGF-BB在不同浓度及孵育时间时可诱导大鼠HSC-T6细胞的增殖,呈现时间依赖性,当剂量到达40 ng/mL时,增殖出现饱和现象;10~40 ng/mL浓度范围内的PDGF-BB孵育12 h和24 h时,可显著升高CollagenⅠ、TGF-β1、TIMP-1的mRNA水平(P<0.05),显著降低MMP-2的mRNA水平(P<0.05),达36 h时,CollagenⅠ的mRNA表达量逐渐下降。结论 10~20 ng/mL浓度范围的PDGF-BB刺激大鼠HSC-T6细胞24 h时对细胞增殖及表达细胞外基质最明显。
Objective To investigate the effects of platelet-derived growth factor-BB in different conditions on the cell proliferation and extracellular matrix secretion in rat hepatic stellate cells T6(HSC-T6)in vitro.Methods HSC-T6 were stimulated with various concentrations(5,10,20,40 ng/mL) of PDGF-BB for different incubation period(3,6,12,24,36 h).The effects of PDGF-BB on cell proliferation were observed by MTT assay method.Real-time PCR was used to measure the mRNA expression of CollagenⅠ,TGF-β1,TIMP-1,and MMP-2.Results Various concentrations of PDGF-BB for different incubation period promoted the HSC-T6 cell proliferation in a time-dependent manner in vitro.But there was a saturation effect at the concentrations of 40 ng/mL.PDGF-BB with concentrations between 10 ng/mL and 40 ng/mL showed significant effects on increasing the expression of CollagenⅠ,TGF-β1,and TIMP-1 while reducing in the expression of MMP-2.The expression of CollagenⅠwas reduced for 36 h of incubation period.Conclusion Stimulating the HSC-T6 with PDGF-BB at the concentrations of 10 ng/mL and 20 ng/mL and incubating for 24 h are the most obvious impacts on the cell proliferation and extracellular matrix secretion in vitro.
