猪源纤维蛋白贴生物相容性初步研究

摘要(Abstract):

目的研究猪源纤维蛋白贴(DBT)生物相容性,为临床应用提供依据。方法通过体外溶血性试验比较不同浓度DBT浸提液对新西兰兔血细胞混悬液中红细胞溶解和凝集的影响。通过皮肤局部刺激性试验研究不同浓度DBT浸提液的皮肤刺激性。结果体外溶血试验结果显示DBT各组不产生溶血。皮肤局部刺激性试验结果显示DBT无明显刺激性,与阴性对照组比较无显著差异(P>0.05)。结论 DBT在体外对兔红细胞未见溶血作用,不引起红细胞凝聚。经DBT处理的皮肤部位大体观察和组织病理学检查均未见明显刺激性,显示DBT的生物相容性良好。
OBJECTIVE Biocompatibility of Porcine Fibrin sealant patch(DBT) is investigated in order to obtain the evidence for new clinical use. METHODS The effects on red blood cell lysis and agglutination in New Zealand rabbit's blood cell suspension by different concentrations of DBT extracts were compared via hemolysis test in vitro and local skin irritation test,the irritation of DBT to rabbit skin was also explored. RESULTS The vitro hemolysis test showed DBT did not cause hemolysis and cell agglutination. Skin irritation test indicated that DBT had no irritation to the local skin and DBT test group had no significant difference with the negative control group(P>0.05). CONCLUSION DBT has no hemolytic effect on rabbit's red blood cells in vitro and don't cause red blood cell aggregation.General observation and histopathological examination of the skin treat with DBT show no obvious irritation,indicating that DBT has good biocompatibility.
