川续断皂苷Ⅵ对大鼠颈总动静脉旁路血栓作用研究

摘要(Abstract):

目的研究川续断皂苷Ⅵ对大鼠颈总动静脉旁路血栓的作用,为临床应用提供实验依据。方法通过测定实验大鼠的血小板最大聚集、TXB_2和6-keto-PGF1α指标以及测定颈总动静脉旁路血栓干湿重探索川续断皂苷Ⅵ抗血栓作用。结果与对照组相比,川续断皂苷Ⅵ组对血栓干湿重均减轻,均具有显著性差异(P<0.01),显著升高大鼠血浆内6-keto-F1α浓度(P<0.05,P<0.01),降低TXB_2浓度(P<0.05,P<0.01),提高6-keto-F1α/TXB_2比值(P<0.05,P<0.01),有效调节体内6-ketoF1α与TXB_2的平衡,抑制血小板的聚集。结论川续断皂苷Ⅵ在预防和治疗血栓中具有一定的活血化瘀,抗血栓作用。
OBJECTIVE To study the effect of asperosaponin Ⅵ on rat common arteriovenous bypass thrombosis and provide experimental evidence for clinical application. METHODS The antithrombotic effect of asperosaponin Ⅵ was explored by measuring the maximum platelet aggregation,TXB_2 and 6-keto-PGF1α indexes,and measuring the wet and dry weight of the common arteriovenous bypass thrombus. RESULTS Compared with the control group,the dry and wet weight of the thrombus were reduced in the group which treated by asperosaponin Ⅵ,there was significant difference(P<0.01).Also the 6-keto-F1α concentration in the rat plasma was significantly increased in the asperosaponin Ⅵ group(P<0.05,P<0.01),TXB_2 concentration was reduced(P<0.05,P<0.01).The ratio of 6-keto-F1α/TXB_2 was increased(P < 0. 05,P < 0. 01). The balance of 6-keto-F1α and TXB_2 were effectively adjusted in vivo and platelet aggregation was inhibited. CONCLUSION Asperosaponin Ⅵ can significantly inhibit thrombosis,which can play a role in prevention and treatment.
