基于聚类分析及主成分分析的猴耳环消炎颗粒指纹图谱研究

摘要(Abstract):

目的建立猴耳环消炎颗粒的HPLC指纹图谱分析方法,并进行聚类分析和主成分分析。方法采用Phenomenex Kinetex C_(18)色谱柱(100 mm×4.6 mm,2.6μm);流动相:乙腈-0.2%磷酸水溶液,梯度洗脱;流速:1.0 m L·min~(-1);柱温为25℃;检测波长:325 nm;测定了两家企业共17批猴耳环消炎颗粒的HPLC图谱。采用中药色谱指纹图谱相似度评价系统(2012年版)及SPSS统计学分析软件进行系统分析。结果 17批猴耳环消炎颗粒的HPLC图谱具有很好的一致性,共标定了7个共有峰,相似度均大于0.9;聚类分析结果可聚成2类;主成分分析结果筛选出4个决定峰。结论所建立的HPLC指纹图谱方法可为全面控制猴耳环消炎颗粒的质量提供依据。
OBJECTIVE To establish HPLC fingerprint of Houerhuan anti-inflammation granule,and provide an evaluation by principal component analysis and cluster analysis.METHODS The 17 batches of Houerhuan anti-inflammation granule were analyzed on a Phenomenex Kinetex C_(18) column(100 mm× 4.6 mm,2.6 μm) with 0.2% phosphoric acid-acetonitrile as the mobile phase for a gradient elution at a flow rate of 1.0 m L·min~(-1).The detection wavelength was 325 nm,column temperature was 25 ℃.RESULTS Seven common peaks were identified in chromatograms with reference to myricetin peak from the 17 batches of the samples.Four main peaks were obtained and all the peaks were classified into two categories.CONCLUSION The developed HPLC fingerprint method could be applied for the quality assessment of Houerhuan anti-inflammation granule effectively.
